Princess Pictures has renewed its relationship with the pair to launch the new studio-based series, Broadcast reports.
The show is reportedly a co-production between Princess and Perkins's indie Square Peg TV, and is being lined up for the 4pm timeslot.
The Great British Bake Off hosts will interview celebrities and real-life characters on the chatshow, which is the pair's first daytime job since they fronted Light Lunch and Late Lunch on Channel 4 between 1997 and 1998.
Helen Warner, ITV's director of Daytime, said: "I've been a big Mel and Sue fan for years. Both are brilliant broadcasters and comedians and bring a unique warmth and charisma as presenters."
Meanwhile, Princess Productions' Emma Hardy added: "We're so excited to be producing this new series for ITV, and thrilled to be making live television again with Mel and Sue...
